Figure 1: Twist Combinatorial TCR Library with 4 defined alpha chains and 4 defined beta chains. Due to our combinatorial assembly, this will yield a library with 16 unique alpha and beta chain combinations. This provides all possible alpha and beta combinations rather than just a subset as described with arrayed libraries.

KEY BENEFITS

High Diversity and Quality

  • Each combinatorial assembly can be composed of up to 10,000 gene fragment combinations
  • NGS-verified libraries with > 90% of possible variants present within 10x of the mean

Customization

  • Customize your TCR Library with user defined combinatorial variants across alpha and beta chains
  • Customize your CAR Library with user defined combinatorial variants across hinge, transmembrane, and signaling domains

Flexibility with throughput

  • Insert sequences up to 1.5 kb in length
  • Libraries designed at scale with diversity across multiple elements of the sequence

Performance Data

The discovery of TCRs is driven by the screening of immune cell repertoires and the re-synthesis of large numbers of potential binders. TCR Libraries can be leveraged to rapidly generate combinations of TCR beta and alpha chains for high throughput screening. With combinatorial TCR libraries, the alpha and beta chains are shuffled together to create a library of greater diversity to integrate the variant space beyond the identified repertoire.

Let’s get Started

The New Digital Library Design (DLD) Tool is your all in one solution to streamline, optimize, and expedite the DNA Library design and ordering process.

Step 1

Log into your account. If you don’t have one, it only takes a few minutes to set it up!

Step 2

Start to design your library in the new interactive libraries design tool which has a step by step “how to” wizard. With this option you can optimize your library in real time with validations on the fly and real-time pricing and receive an SOW at the end of the submission process.

Step 3

Once the quote is accepted by the customer, the Libraries Team will send the project to the production team

Note:

The Libraries Design Tool is not currently available in the APAC region. Please check with your local support team on availability date.
